Wade Wilson is a former spetsnaz soldier working as a mercenary in New York . In the local bar of his friend Ferdinand Wade meets a prostitute Vanessa Carlisle, with whom he later has a romantic relationship. A year later, Wade makes a proposal to Vanessa, and she agrees. Suddenly Wade faints and the next day he is diagnosed with cancer in the last stage. Vanessa asks Wade to stay with her, but Wade does not want her to see his death.

In the Ferry bar, Wade is approached by a recruiter from a secret program offering experimental cancer treatment. For Vanessa Wade agrees to experiments. There he meets the head of the program, a former informer in the laboratory of Dr. Killebrew, a mutant Ajax and his henchman, a participant in the experiments Weapon X Christina, nicknamed Angelic Dust. Ajax introduces Wilson’s serum-mutagen, designed to activate the DNA mutationrights. To activate the serum, adrenaline is needed, and Wade is subjected to various kinds of monstrous tortures that do not bring success. In the process, Wade recognizes the real name of Ajax (Francis Freeman) and teases him. Before that, Ajax reveals to Wade the true goal of the program: to empower people with superpowers and sell them into slavery. During the torture, Wade’s mutation is activated, and he receives a healing factor, due to which the cancer does not have time to form. However, while Wade gets a side effect – his body and face are disfigured. Wade manages to make an explosion in the laboratory, firing a fire to supply oxygen in the cell, so that he is released and enters into a fight with Ajax. In the process, Ajax reveals to Wade that he alone can restore his previous appearance, then pierces Wilson with steel reinforcement and leaves to die in a burning laboratory. The lab is completely burned, but Wade is getting out from under her remains.
Wade does not dare to return to Vanessa because of the disfigured appearance. After a conversation with Ferret, Wade decides to find Ajax and regain his former appearance. He becomes a disguised lynch player and takes the name “Deadpool” (from Dead Pool – the deadly tote in the Ferry bar). Wade settles in the house with a blind elderly woman Al. The Colossus and the Warhead enter battle with Angelic dust, while Deadpool pushes the way to Ajax through his assistants.
With the help of the Warhead, Deadpool pierces the helicopter and engages in a fight with Ajax. In the meantime, saving the Colossus, the Warhead accidentally destroys the helicarrier supports, and it begins to fall. The Colossus takes the Warhead and the Angelic dust to a safe place, and Deadpool saves Vanessa. Ajax survives and attacks Deadpool, but is defeated. He reveals that Deadpool’s appearance can not be remedied, and Wade, despite Coloss’s requests to keep him alive, kills Ajax. Vanessa is angry at Wade because he left, but forgives him. Wade shows her a disfigured face, but she accepts it, and they are reunited.
In the scene after the credits, Deadpool appears in the dressing gown and reveals that in the sequel of “Deadpool” will appear Cable
